چکیده

The identification of eight previously undescribed polypeptides in chicken embryo cells infected with Sindbis virus is reported. Seven of these polypeptides were distinguishable from the virus structural polypeptides and their precursors by their molecular weights and tryptic peptide maps. The eighth was closely related to pE2 (Schlesinger and Schlesinger, 1973), a precursor to one of the virus particle glycoproteins. Pulse-chase experiments and the use of an inhibitor of proteolytic cleavage allowed a division of the seven nonstructural (NS) polypeptides into three stable end products (NS p89, NS p82, and NS p60) and four precursors (p230, p215, p150, and p76). The labeling kinetics after synchronous initiation of translation indicated that synthesis of the NS polypeptides started at a single site and showed that the order of the genes coding for the NS polypeptides was (5' -+ 3') NS p60, NS p89, and NS p82. Short-pulse experiments under conditions of both synchronized and nonsynchronized translation suggested that cleavage of the primary translation product ofthe NS genes occurred only after its synthesis was completed and that the first cleavage removed the Cterminal polypeptide. From these and other experiments, we propose a detailed scheme for the synthesis and processing of Sindbis virus NS polypeptides.

Characterization of the binding of multiplication-stimulating activity to a receptor for growth polypeptides in chick embryo fibroblasts.

Multiplication-stimulating activity (MSA) is the name given to a family of polypeptides with a molecular weight of approximately 10,000 that have been isolated from the culture medium of a rat liver cell line. MSA stimulates growth and DNA synthesis in chick embryo fibroblasts and has weak insulin-like metabolic activity. We have purified MSA and characterized the binding of ‘““I-labeled MSA to...

Repression of quiescence-specific polypeptides in chicken heart mesenchymal cells transformed by Rous sarcoma virus.

Chicken heart mesenchymal cells do not proliferate in medium of physiological composition containing plasma (S. Balk, Proc. Natl. Acad. Sci. USA 77:6606-6610, 1980). To understand the molecular events involved in cell quiescence and in the initiation of cell division under physiological conditions, we examined the differences in the patterns of protein synthesis of quiescent, hormone-stimulated...

VP5, the nonstructural polypeptide of infectious bursal disease virus, accumulates within the host plasma membrane and induces cell lysis.

Infectious bursal disease virus (IBDV) encodes a 17-kDa nonstructural polypeptide known as VP5. This polypeptide is not essential for virus replication in vitro but it plays an important role in in vivo dissemination and pathogenesis. We have characterized the expression of VP5 in three eukaryotic systems: (i) IBDV-infected chicken embryo fibroblasts; (ii) BSC-1 cells infected with a recombinan...

Identification of somatomedin-like polypeptides produced by mammary tumors of BALB/c mice.

A transplantable mammary tumor derived from an outgrowth of nodule-like alveolar lesions induced by 7,12-dimethylbenz[a]anthracene in cultures of whole mammary gland was shown to produce a family of somatomedin-like polypeptides when cultured in vitro. Minced mammary tumor tissue as well as monolayer cultures of tumor cells produced similar polypeptides when incubated in serum- and hormone-free...

Induction of virus-neutralizing antibody by glycoproteins isolated from chicken cells infected with a herpesvirus of turkeys.

The production of virus-induced proteins in chicken embryo fibroblast cells infected with a herpesvirus of turkeys was studied. It was found that glycoproteins isolated from membrane-rich fractions of infected cells by affinity chromatography using concanavalin A induced neutralizing and precipitating antibody in rabbits and chickens. After analytical electrophoresis, such isolates were found t...
